NgAgo 或只能切割 RNA

1月20日，美國冷泉港實驗室（CSHL）旗下的預印本網站 BioRxiv 發表了一篇來自韓國基礎科學研究院基因組工程中心、科學技術聯合大學院和韓國國立首爾大學化學系Jin-Soo Kim課題組的題為「DNA-dependent RNA cleavage by the Natronobacterium gregoryi Argonaute」的文章，該文指出NgAgo介導的DNA依賴的RNA切割是有效的。

值得注意的是該文的通訊作者Jin-Soo Kim正是2016年11月28日領銜在Nature Biotechnology雜誌發表題為「Failure to detect DNA-guided genome editing using Natronobacterium gregoryi Argonaute」的 correspondence 類型的文章，直指 NgAgo 不能夠進行基因編輯。Jin-Soo Kim 一直是基因編輯領域的大腕，在 NBT 和其他一流雜誌上曾發表過多篇關於基因編輯的重要文章。

Jin-Soo Kim課題組在預印本網站 BioRxiv 發表的文章在摘要中清晰的指出，NgAgo 能夠作為一種 DNA 引導的核酸內切酶靶向性切割 RNA 而不是 DNA（下圖）。作者證實了 NgAgo 能在體外與5』 羥基化或者磷酸化的單鏈DNA 組合，將 RNA 切割成兩個或多個片段。

NgAgo 在 gODN 的作用下能夠特異性的切割對應基因的 RNA 轉錄本，顯然並不作用於單鏈 DNA（ssDNA）或者雙鏈 DNA（dsDNA）（下圖）。

上述結果很好的解釋了為什麼此前許多實驗室都能夠做出韓春雨去年5月2號發表在 NBT 中的 Figure 3圖，既降低外源 GFP 的表達。這也解釋了去年11月11日Cell Research以Letter to Editor 形式在線發表的來自南通大學和復旦大學研究人員合作完成的題為「NgAgo-based fabp11a gene knockdown causes eye developmental defects in zebrafish」的論文中發現gDNA/NgAgo 系統在斑馬魚中能實現特定基因敲低導致魚眼部發育缺陷，但是不能對靶基因進行基因編輯。此外，更早以前北大魏文勝實驗室運用gDNA/NgAgo 系統也能看到目的基因的表達下調但不能實現DNA編輯。韓國科學家的這項研究可能為gDNA/NgAgo 系統的基因編輯引發的系列爭議給出了比較好的解釋和實驗證據，看來gDNA/NgAgo 系統確實能在體外很好的實現RNA的編輯（下圖）。

關於在8月8日，韓春雨向質粒共享信息庫 Addgene 提交新版的詳細實驗方法，其中特別提到了轉染中應避免 EDTA 對鎂離子的鰲合問題（Because Agos need magnesium, avoid EDTA when detaching and seeding cells into the plates for transfection，引自韓春雨向Addgene提交的新版實驗方法）。在預印本的這篇文章中專門設計了實驗證明在體外實驗中，在有 EDTA 的情況下，NgAgo 確實不能切割 RNA（下圖）。

比較好玩的一點是，NgAgo 能重複使用，反覆切割。相比之下一個 Cas9 分子只能使用一次（下圖）。

作者在文章最後的討論部分提到，NgAgo 系統可以用來作為生物醫學研究的一種 RNAi 工具來介導特異性的 mRNA 或者 lncRNAs 的降解。

要點：該課題組可能為了搶速度，直接把論文公布在預印本網站上而不是走同行評議渠道發表在正式的期刊上，關鍵一點是上述實驗都是在體外進行的，所以後續還需要進一步在體內進行驗證。鑒於課題面臨的競爭，想必還有其他課題組也在完成類似的工作。讓我們拭目以待吧！

【摘要】We show here that, unlike most other prokaryotic Argonaute (Ago) proteins, which are DNA-guided endonucleases, the Natronobacterium gregoryi-derived Ago (NgAgo) can function as a DNA-guided endoribonuclease, cleaving RNA, rather than DNA, in a targeted manner. The NgAgo protein, in complex with 5′-hydroxylated or 5′-phosphrylated oligodeoxyribonucleotides (ODNs) of variable lengths, split RNA targets into two or more fragments in vitro, suggesting its physiological role in bacteria and demonstrating a potential for degrading RNA molecules such as mRNA or lncRNA in eukaryotic cells for study of their functions.

Jin-Soo Kimis an entrepreneur and chemist-turned-biologist. He graduated from Seoul National University in 1987 with a major in chemistry. He then earned a master』s degree in chemistry fromSeoul National Universityin 1989 and a Ph.D. in biochemistry from the University of Wisconsin-Madison in 1994. After postdoctoral training atHoward Hughes Medical Institute/Massachusetts Institute of Technology, he came back to Seoul in 1997 to serve as Principal Investigator at Samsung Biomedical Research Institute. He co-founded a biotechnology company, ToolGen, Inc., focused on zinc finger technology in 1999, and served as CEO and CSO for the subsequent 6 years. He joined the faculty of the Department of Chemistry at Seoul National University in 2005. He now serves asDirector of Center for Genome Engineeringat Institute for Basic Science. He has published over 60 articles and filed 20 patent applications, mostly in the field of gene regulation and genome editing. He has been a member of Faculty of 1000 since May, 2013.
