對流感病毒H1N1有抑制作用的二萜生物鹼

烏頭Aconitum tanguticum為毛茛科烏頭屬植物,主要分布於西藏、青海、甘肅、四川、雲南等省,是一種傳統的中藥,有回陽、逐冷、祛風濕的作用。近日,中國中醫科學院隋峰研究員報道從一種烏頭植物全株中一共分離得到了5個新的二萜生物鹼和2個新的醯胺類化合物,並測試了部分化合物對H1N1的抑制活性。相關研究發表於Phytochemistry,題為 「Diterpenoid alkaloids from the whole plant ofAconitum tanguticum(Maxim.) Stapf」。

化合物1、4、5和7為白色無定形粉末,化合物2(MeOH)和化合物3為無色針狀晶體(CHCl3),化合物6為橘黃針狀晶體(CHCl3)。
利用前期文獻給出的CPE法,測試了化合物1和化合物5對甲型H1N1流感病毒的體外抗病毒活性。兩者對H1N1引起的細胞病變均表現出明顯的抑制作用,IC50分別為2.9μg/mL(TI 3.81)和2.4μg/mL(TI 9.44),而利巴韋林(陽性對照)的IC50為220μg/mL(TI 16.1)。
Abstract
Five undescribed diterpenoid alkaloids tanguticulines A-E and two undescribed amide compounds 5-methoxy-N-Salicylanthranilic acid methyl ester and 3, 5-dimethoxy-4- hydroxycinnamamide-4-O-β-D-glucopyranoside, were isolated from the whole plant ofAconitumtanguticum (Maxim.) Stapf. Their structures were assigned by detailed analysis of MS and NMR spectroscopic data. Compounds1and5were evaluated for their antivirus activities against influenza a (H1N1) virusin vitro. Both of them showed obvious inhibitory effect on the cytopathic changes induced by H1N1, with IC50values of 2.9?μg?mL?1and 2.4?μg?mL?1, respectively.
